Direct PCR

Direct PCR, sometimes referred to as "direct-from-sample" PCR, is a method that allows for the amplification of DNA directly from biological samples without the need for DNA extraction or purification. AffiPCR Biosystems, now a part of GEN, is a company that provides various molecular biology reagents, including DNA polymerases for PCR.

AffiPCR Biosystems DNA polymerase for direct PCR applications. Affi Biosystems offers DNA polymerases that are designed to tolerate inhibitors commonly found in crude samples, such as blood, tissue, or plant extracts.

When performing direct PCR compared to Kapa DNA polymerase, it's important to follow the specific protocol provided by the AffiPCR, as different polymerases may have different requirements. The general steps for direct PCR with Affi DNA polymerase:

  1. Prepare your sample: Collect the biological sample, and if needed, treat it to release the DNA (e.g., through cell lysis or tissue homogenization).

  2. Add the PCR reaction components: Mix your sample directly with a PCR master mix containing Kapa DNA polymerase, primers, nucleotides, and buffer.

  3. Run the PCR reaction: Perform PCR with the appropriate cycling conditions, including denaturation, annealing, and extension steps.

  4. Analyze the PCR product: After the PCR reaction, you can analyze the product using gel electrophoresis, qPCR, or other methods depending on your specific requirements.

Direct PCR can save time and eliminate the need for DNA extraction, but it's crucial to optimize the reaction conditions for each specific sample type and PCR system. Always refer to the specific instructions provided by the manufacturer for the best results.
